Mesenchymal stromal cells induce MYC repression in activated T cells

We utilized 10X Genomics Multiome technology to obtain single cell gene expression and chromatin accessibility profiles to identify the genetic and epigenetic changes which underlie the suppressive effects of yMSCs on alloreactive T cells. Overall design: Naïve T cells from 5 anonymous human donors activated for 6 days by allogeneic DCs from a sixth donor. Cells in experiemental group exposed to yMSC on the bottom of a transwell insert for 48 hours before being placed back into fresh media. snRNAseq and snATACseq libraries generated with 10x Multiome at every timepoint for longitudinal computational analysis.
