Hepa1-6 cells were subcutaneously injected into the right hind limbs (defined as Tumor1) and left forelimb armpit (defined as Tumor2) of C57BL/6 mice respectively. Seven days after the inoculation, the tumors were established. For evaluating the efficacy of combing poly(I:C) with RT, the bilateral HCC subcutaneous tumor bearing mice were randomly divided into four groups, including normal saline (NS) treated group, poly(I:C) treated group, RT treated group, and poly(I:C) plus RT treated group. The mice were administered with poly(I:C) (200 µg per mouse) or NS (200 µl per mouse) by subcutaneous injection at the day1, day3 and day5~day11 of treatment procedure. In the experimental group that received RT, the tumor1 in tumor-bearing mice was irradiated with a total dose of 40 Gy (8Gy*5days). Total RNA of C57BL/6 mouse tumor tissues (3 tissues/group) was extracted. RNA library sequencing was performed by Berry Genomics on illumina Novoseq 6000 (paired end, 150bp).
