Growth curve of algae and betaine content determination

Betaine and carnitine standard/Sample solutions were derivatised with a mixture of 2-Bromo-2'-acetonaphthone and 18-crown-6 in N-methyl -2 pyrrolidone in presence of KH2PO4 (0.1 M) at 80◦C (4 h). After filtering, the solution was eluted with 22 mM Choline chloride in 10% H2O/90% ACN at a flow rate of 1.9 ml/min (Injection 5 µL) at 40◦C. Betaine content in microalgae biomass was determined using calibration curve of standard betaine.

标准甘氨酸和肉碱样品溶液在磷酸二氢钾（0.1 M）的存在下，于80℃（4小时）与2-溴-2'-乙酰萘酮和18-冠-6的混合物在N-甲基-2-吡咯烷酮中进行衍生化处理。过滤后，溶液以1.9 ml/min的流速用22 mM的胆碱氯化物在10%水/90%乙腈中洗脱，注射量为5 µL，在40℃下进行。通过标准甘氨酸的校准曲线确定微藻生物量中的甘氨酸含量。
