During activation T cells need to undergo metabolic reprogramming for proper effector function. Disturbances in metabolic pathways lead to impaired T cell function. Active metabolic evasion of cancer cells lead to less immunosurveillance and uncontrolled tumor growth. We used microarrays to investigate the expression changes in T cells challenged with AML supernatants shedding light on the underlying mechanisms of metabolic evasion in leukemia. Primary murine T cells isolated from the spleen were activated and cultured in fresh medium (Tc.Tc) as a control, medium conditioned by WEHI 3B murine AML cells (Tc.WEHI) or bone marrow derived dendritic cells (Tc.Dc). CD8+ T cells were reisolated after 48 hrs for RNA extraction and hybridization on Affymetrix microarrays. Seven biological replicates per condition were analyzed.
