This project includes raw sequencing data from a genome-wide transposon sequencing (Tn-seq) experiments performed on Neisseria mucosa, a prevalent oral commensal increasingly associated with chronic respiratory diseases. The goal was to identify genetic determinants that support N. mucosa fitness under conditions mimicking the inflamed lower away. Transposon mutant libraries were grown under the following conditions: (1) Input: Frozen library samples revived in nutrient-rich media, representing baseline mutant abundance; (2) SCFM2 (anoxic): Synthetic Cystic Fibrosis Medium 2, a chemically defined medium simulating sputum from individuals with bronchiectasis; (3) TSBYE + nitrate (anoxic): nitrate-supplemented standard laboratory media (tryptic soy broth + yeast extract) incubated under anoxic conditions; (4) TSBYE (oxic): same media without nitrate incubated under oxic conditions. Libraries were sequenced to assess mutant fitness across conditions. Data provide insights into N. mucosa metabolism, antibiotic resistance and potential vulnerabilities that may be targeted in the context of chronic airway disease.
