We studied how H1 regulates higher-order chromatin architecture by performing Hi-C. In addition, we tested the effects of H1 domains on nucleosome organisation and DNA methylation by performing MNase-seq and Bisulfite-seq. Overall design: Hi-C was performed with h1 mutant seedlings to examine chromatin interactions in the mutant. To test the functions of the C-terminal intrinsically disordered region (IDR) of H1, we generated H1.1G18IDR which contains the IDR and 18 amino acids of the globular domain nearest the IDR in h1 background. As a control, H1.2 with IDR deleted (H1.2?IDR) in h1 background was also generated. These transgenic plants as well as WT and h1 mutant were subject to MNase-seq and Bisulfite-seq to analyse nucleosome repeat length and DNA methylation.
