Transcriptional control of dendritic cell (DC) homeostasis has not been fully understood. Genome-wide binding analysis of TRIM33 in dendritic cells revealed a critical role in gene regulation for DC differentiation. Overall design: To determine the genome-wide binding sites of TRIM33 in DCs, chromatin immunoprecipitation-sequencing (ChIP-seq) assays were performed. An immortalized cell line named Mutu DC1940 (Marraco et al., 2012) was found to harbor much primary DC features, and is therefore suitable for sampling. 2 biological repeats of TRIM33-immunoprecipitated chromatin and the corresponding input and IgG controls were prepared from wildtype Mutu DC1940 cells. The Illumina NovaSeq 6000 platform was used to generate PE150 reads.
