Chimeric antigen receptor (CAR) T cells have become powerful players in immune-based cancer therapy space, but current lentiviral and retroviral-based production systems are limited by cost and modularity. Improved systems for efficient generation and enhanced features are therefore of great interest. Here we developed a method using the CRISPR-Cpf1 system to generate a stable CAR-T based on homology-directed repair (HDR) knock-in and immune checkpoint knockout in one step.
