Exceptional preservation of DNA in ancient permafrost offers a unique opportunity to examine fossil and living microorganisms over geological timescales. DNA repair protocols were combined with high throughput sequencing to construct metagenome-assembled genomes from ancient microbial DNA entrapped in Siberian coastal permafrost with freezing over 100 kyr ago.
