High-fidelity target sequencing of individual molecules identified by barcode sequences. Homo sapiens

Labeling DNA fragments with barcode sequences is a promising approach in the high-fidelity sequencing of single molecules with massively parallel sequencers. However, the basic principle of labeling each molecule with a single unique barcode is unworkable due to the high read error rate of current sequencing technology. We developed a method that can be used to monitor and remove erroneous barcode sequences using a novel target sequencing method adding barcode sequences by adaptor ligation.
