Our study focuses on gliomas, the most common primary brain tumors in adults, and understanding the role of endogenous transposable elements in these malignancies. Using genomic DNA from 7 low passage oncosphere cell lines derived from primary GBM biopsies, 3 secondary GBM tissue samples, and matched normal blood samples, we performed transposon insertion profiling by deep sequencing (TIPseq) of the active subfamily of Long INterspered Element-1 (LINE-1) elements to find evidence of somatic retrotransposition. Our findings indicate that although LINE-1 can act as an infrequent insertional mutagen in secondary glioblastomas, retrotransposition is uncommon in these central nervous system tumors as compared to other neoplasias.
