Chlamydia trachomatis serovar D plaque purified strains were passaged in the presence of the HtrA inhibitor compound JO146 to select for resistant mutants. 3 strains that were least susceptible to JO146 were selected for whole genome sequencing. 2 of the 3 strains were mutated using ethyl methanesulfonate (EMS) prior to passaging in the presence of JO146.
