The proportion of pathogenic Th17 (pTh17) cells were significantly higher in RA than that in control individuals and showed a potential relevance with YY1 expression. Increased YY1 expression was observed in pTh17, and the pTh17 differentiation was hampered by YY1 knockdown. We used microarrays to detail the gene expression changes of pTh17 transfected with YY1 shRNA lentivirus and tried to elucidate the potential mechanisms how YY1 can affect pTh17 cell differentiation in RA. Human naïve CD4+ T cells from peripheral blood mononuclear cells of 3 healthy honors were purified and only a CD4+CD45RA+ cell suspension with a purity ≥ 90% was used for following experiments. Purified naïve CD4+ T cells at 2×105 cells per well were cultured in 96-well round-bottom plates with the following antibodies and cytokines for pTh17 subset polarizing condition in RPMI-1640 with 10% fetal bovine serum(FBS): 2 μg/ml anti-CD3/CD28, 100 U/ml IL-2, 10 ng/ml IL-6, 10 ng/ml IL-1β, 10 ng/ml IL-23, 5 μg/ml anti-IL-4, 5 μg/ml anti-IFN-γ. The polarized pTh17 subsets were cultured with YY1 shRNA lentivirus (MOI=20), control lentivirus (MOI=20) after cytokines stimulation for 24 h .Cells were grown for 3 days at 37°C in 5% CO2. On day 4, cells were extracted using the standard Trizol method for microarray.
