In this study, populations of knockout cells were generated using the Yusa v3 or Horizon sgRNA libraries in 6 cell lines: CAMA1, U2OS, DLD1, T47D, HCC1806 and BT549. Populations of knockout cells were treated with the PARP inhibitor olaparib for 21 days. Over this time, knockout cells that were sensitive to olaparib treatment would die and a population of knockout cells that was resistant to olaparib treatment remained. Cells from olaparib and DMSO conditions and the sgRNA cassette in the knockout cells were amplified and sequenced to identify synthetic lethal interactions with olaparib treatment.
