Our findings demonstrate that XBP1 controls macrophage cGAS/STING/NLRP3 activation by regulating macrophage self-mtDNA cytosolic leakage via BNIP3-mediated mitophagy modulation, providing a novel target against liver fibrosis. Overall design: BMDMs were isolated from Xbp1FL/FL and Xbp1M-KO mice and lysed in TRIzol reagent (Invitrogen, Carlsbad, CA) after LPS stimulation for 6 h, and then Xbp1FL/FL and Xbp1M-KO BMDMs were subjected to RNA sequencing analysis (n = 3/group).
