We used a CRISPR-based screening approach to identify functional vulnerabilities in pediatric sarcoma cell types, identifying BRD9 as an important dependency in synovial sarcoma cells. Overall design: ChIP-seq of BRD9, SS18-SSX1 in cells treated with BRD9 targeting degraders. ChIP-seq of steady state profiles of H3K27Ac and RNAPII. ChIP-seq of WT and mutant BRD9 proteins at steady state.
