The primary objective of this study is to conduct a comprehensive transcriptomic analysis of mouse MH-S alveolar macrophages after exposure to Aspergillus fumigatus culture supernatant. This approach aims to elucidate the global changes in gene expression underlying the initial immune response of alveolar macrophages (key sentinel cells in the lung) to the soluble mediators and antigens secreted by this ubiquitous fungal pathogen. By focusing on fungal supernatants, which comprise complex mixtures of metabolites, enzymes, and cell wall components, we aim to identify specific signaling pathways, inflammatory cascades, and cellular processes, such as cytokine production, phagocytosis, and oxidative stress, that are either activated or suppressed. The relevance of this study is critical, as Aspergillus fumigatus is a major opportunistic fungus responsible for a range of respiratory diseases, from allergic aspergillosis to life-threatening invasive pulmonary aspergillosis, particularly in immunocompromised individuals. Understanding the precise molecular dialogue between host frontline defense cells and fungal secreted factors is essential for elucidating the pathogenesis of Aspergillus-related diseases. Ultimately, the findings of this study are expected to reveal novel host-pathogen interactions and identify potential molecular targets, providing a foundation for the development of innovative diagnostic and therapeutic strategies against Aspergillus fumigatus infections.
