Comprehensive synthetic library to untangle genetic and phenotypic determinant of translation efficiency. Escherichia coli strain:MDS42 recA

A precisely designed synthetic library comprising 244,000 variants of a 96 nucleotide sequence fused to a fluorescent reporter was build to unravel the determinant of translation efficiency in E. coli. Combinations of key sequence properties are systematically implemented. Multiple phenotypic consequences of the sequence perturbations are measured using high-throughput sequencing : RNA abundance and decay, ribosome densities, protein production and growth rate.
