Neutrophil recruitment appears critical to the pulmonary defense to B. pseudomallei. Therefore, we hypothesized that ?d T cells are critical regulators of pulmonary bacterial clearance given their proposed regulation of neutrophil recruitment to the lung. To investigate this hypothesis, we leveraged multiple in vivo models of pulmonary melioidosis to further understand the mechanisms by which ?d T cells regulate inflammation early in severe pulmonary infection. Included in this investigation, we assessed whole lung bulk RNA-sequencing data in mice with and without ?d T cells after infection with Burkholderia thailandensis. Overall design: We infected 8-12 week old female wild type C57BL/6 mice and Tcrd-/- mice on a C57BL/6 background with whole body aerolozide B. thailandensis. At 24 hours after infection, we procured whole lungs, extracted RNA and then performed bulk RNA-sequencing. Uninfected control mice of both backgrounds also had their whole lungs procured, RNA extracted and bulk RNA-sequencing performed concurrently.
