CRISPR knock-out screen for MEFV M694V inflammasome-mediated cell death

The MEFV M694V mutations is associated with the pediatric autoinflammatory disease Familial Mediterranean Fever syndrome. MEFV M694V encodes constitutive active PYRIN protein that induced inflammasome activation and inflammation wihtout need for activation signal. Inflammasome activation leads to release of cytokines and inflammatory cell death (pyroptosis). This screen aimed to identify positive regulators of the inflammasome pathway induced by constitutive active PYRIN.
