A DNA mixing pool was constructed by polling the DNA samples collected from 26 purple- (Rpool) and green-leaved (Gpool) F2 plants. Four samples of Gpool, Rpool, and two parental lines, Gp and Rp, were used for paired-end (150 bp) sequencing using the Illumina HiSeq platform. In the F2 generation, two populations with extreme traits were selected to construct gene sequencing mixing pools for bulked segregant analysis sequencing. Thirty-nine genes controlling leaf color were localized to a 200 kb region on chromosome 7 using combined delta (SNP-index) and delta (Indel-index) analyses.
