Epigenomic profiles of 4T1 cell line in primary, metastasis site and circulating cell type in xenograft mouse model

We developed the platform for harvesting CTCs by modifying our previously-developed microfluidic-based CTC capturing system. We successfully cultivated and expanded the CTCs for methylomic analyses based on the high efficiency and excellent viability of this modified CTC-capturing system.About 10e5 4T1-hHER2-Luc cells was orthotopically inoculated into fat pad of the NOD-SCID-gamma mice. The primary tumor and second metastasis lung tissue were collected after 4 weeks of cancer growth and progression. Whole blood of the mice were collected for CTC isolation. the 4T1-derived primary, CTC, and secondary metastasis cancer cell lines were utilized for bisulfite sequencing.
