RNAseq analysis of microglia isolated from treated brain slices or treated mice

Population based transcriptomic profiling was performed on microglia isolated from TNF or GMCSF treated brain slices or from LPS treated mice Overall design: For organotypic brain slice microglia samples, brain slices were cultured for 2 weeks and then treated for 24 hours with TNF (50 ng/mL), or GMCSF (20 ng/mL). Following treatment microglia were isolated for analysis, to gain sufficient RNA material for analysis each sample consists of 3 pooled brain slices. For microglia derived from LPS treated mice, mice were treated with LPS (1 mg/kg) injected intraperitoneally 24 hours prior to microglia isolation. In the case of brain tissue analysis samples were flash frozen in liquid nitrogen at the time of collection.
