资源简介:
The goal of this Whole Genome Sequencing (WGS) analysis was to identify genes underlying TCS in C. elegans strain expressing the gut-specific hsp-90 hairpin RNAi construct compared to a control strain (strain AM994). To do this we performed a forward genetic screen using the mutagen EMS and screened progeny for reduced TCS-mediated expression of the hsp-70p::mCherry reporter in muscle cells. The forward genetic mutagenesis screen identified four mutants showing reduced hsp-70p::mCherry expression. These mutants were taken forward to a one-step WGS and SNP mapping approach as described in Doitsidou M, Poole RJ, Sarin S, Bigelow H, Hobert O. C. elegans mutant identification with a one-step whole-genome-sequencing and SNP mapping strategy. PLoS One. 2010;5(11):e15435. Published 2010 Nov 8. doi:10.1371/journal.pone.0015435.
