Here, we report a virus-like recombinant biosome (reBiosome) as a safe and effective delivery vehicle for targeted gene therapy. A mutant biosome (mBiosome) was first prepared by genetically recombining a vesicular virus glycoprotein (VSV-G) on a natural biosome by rationally displaying an unnatural amino acid, and the reBiosome was then prepared by chemical addition of a stealth polymer material onto the mBiosome. The results showed that the reBiosome could target weakly acidic foci and had a high gene transfection efficiency with a reduced virus-like pro-inflammatory effect, demonstrating robust therapeutic efficacy in gene editing therapy of breast cancer and in gene silencing therapy of arthritis. In conclusion, this study offers a novel virus-like recombinant biosome that enables targeted gene therapy.
