In this study, we reporte the altered miRNAs expression in the human triple negative breast cancer cell line (MDA-MB-231) in response to withaferin A at IC50 (2 µM) concentration. Withaferin A is a major pharmacological active constituent of Withania somnifera (Solanaceae) plant. In this study we were aimed to identify the diffentially expressed known/novel miRNAs using next generation sequencing (NGS) in withaferin A treated MDA-MB-231 cells. For this, Human breast TNBC (MDA-MB-231) cells were cultured in the Dulbecco’s Modified Eagle Medium (DMEM) media supplemented with 10% (v/v) heat-inactivated FBS, 100 U/ml of penicillin, and 100 mg/ml of streptomycin and maintained in 5% CO2 at 37 degre celcius in a humidified atmosphere of 95% air in CO2 incubator. A total of 1x105 MDA-MB-231 cells were seeded in 35mm treated culture dishes containing 1ml of DMEM culture media and sub-cultured at every 3–4 days. The cultured MDA-MB-231 cells were treated with WA (2 µM) for 24 h as experimental group and without WA treated MDA-MB-231 cells were taken as control group. Three samples were assigned for each group.
