This dataset supports my thesis on the development and preclinical evaluation of novel DelNS1 live attenuated influenza vaccines (LAIVs). The research scope encompasses two vaccine candidates: (1) a trivalent DelNS1 LAIV targeting the 2020–2021 seasonal influenza strains (A/H1N1, A/H3N2, and B/Victoria), and (2) a monovalent DelNS1 LAIV against highly pathogenic avian influenza (HPAI) H5N1 (clade 2.3.4.4b). Using BALB/c mice and hamsters as animal models, I assessed immunogenicity, protective efficacy, and the impact of different administration routes (intranasal, intramuscular, and heterologous prime-boost regimens). Key findings include that a single intranasal dose of the trivalent vaccine induced potent serum neutralizing antibodies, robust mucosal IgA responses in the respiratory tract, and tissue-resident memory CD4+/CD8+ T cells in the lungs. The monovalent H5N1 vaccine provided sterilizing and long-lasting immunity against lethal challenge with heterologous HPAI H5N1 viruses. Additionally, intranasal vaccination stimulated immune responses in the small intestine, suggesting broader mucosal vaccine applications.The dataset is provided as a single Excel file containing all raw data presented in my thesis. Each worksheet is clearly labeled by the corresponding thesis chapter (e.g., Chapter 3), and includes measurements such as viral titers, body weight changes over time, serum IgG antibody titers, mucosal IgA levels, and influenza-specific CD4+ or CD8+ T cell frequencies.
