Mechanism of TCONS_00147848 regulating apoptosis of nasal mucosa cells and alleviating allergic rhinitis through FOSL2-mediated JAK/STAT3 signaling pathway

This study aims to explore the roles, and related mechanisms of TCONS_00147848 in nasal mucosa cells apoptosis and allergic rhinitis (AR). The AR mice were sensitized with ovalbumin (OVA), and the lentiviral expression vector of TCONS_00147848 shRNA was constructed. NC shRNA and TCONS_00147848 shRNA were transfected into AR mice and mice nasal mucosa cells induced by TNFa. The allergic reaction symptoms were evaluated by scoring. HE staining and TUNEL were used to detect the histological changes and apoptosis of nasal mucosa in mice. CCK-8 assay, Transwell and annexin VPI were used to detect proliferation, migration and apoptosis of mice nasal mucosa cells, respectively. ELISA was used to detect the expression of inflammatory factors. The expression of TCONS_00147848 was detected by qRT-PCR. Western blot assay was used to detect the expression of FOSL2, JAK-2, STAT3, p-STAT3, BAX and BCL-2. RNA-binding protein immunoprecipitation (RIP) assay, RNA pull down assay and Co-immunoprecipitation (CoIP) assay were applied to identify TCONS_00147848 that target FOSL2.
