Purpose: This study uses transcriptome analysis (RNA-seq) to study the effects of photobiomodulation therapy on classically activated M1 macrophages induced by LPS and IFN-?. Overall design: After 12 hours of LPS and IFN-? induction, the differentially expressed genes of classically activated macrophages in the blank control group and the PBM treatment group were examined. Total RNA was isolated from macrophage sample using the standard TRIzol protocol (Invitrogen, Carlsbad, CA, USA). Strand-specific libraries were constructed using the TruSeq RNA sample preparation kit (Illumina, San Diego, CA, USA), and sequencing was carried out using the Illumina HiSeq X Ten instrument by the commercial service of Genergy Biotechnology Co. Ltd. (Shanghai, China).
