A549细胞系来源于一位58岁肺癌男性患者,由D.J. Giard等人于1972年通过组织原代培养获得。M. Lieber等人的进一步研究表明A549细胞系能够通过胞苷二磷酸胆碱途径借助高不饱和脂肪酸合成卵磷脂,且该细胞免疫酶标染色呈现角蛋白阳性反应。
无胸腺裸鼠(Nude Mouse,简称裸鼠)目前已成为医学生物学研究领域中不可缺少的实验动物模型。经过世界各国实验动物遗传育种学家的努力,目前已将nu基因导入不同近交系动物,成为系列动物模型,仅小鼠模型一种,已建立了二十余种近交系裸鼠。由于裸鼠的免疫缺陷,在一定情况下,不排斥来自异种动物的组织移植。因此可作移植人类恶性肿瘤的接受体。根据T.Fogh等1979~1980年的报导,已有150株人的瘤细胞和人体原发癌移植于裸鼠获得成功,目前已成功地将人结肠癌、乳腺癌、肺癌、卵巢癌、黑色素瘤、淋巴瘤、白血病、肾癌、宫颈癌、软组织肉瘤和骨肉瘤等移植于裸鼠、获得了一定百分比(35.7%)的良好生长,并可传代。若用已建株的人体肿瘤组织培养细胞作移植材料,接种后的成活率更高(41%)。
The A549 cell line was derived from a 58-year-old male lung cancer patient, and was established via primary tissue culture by D.J. Giard et al. in 1972. Further studies by M. Lieber et al. demonstrated that the A549 cell line can synthesize lecithin via the cytidine diphosphate choline pathway using highly unsaturated fatty acids, and its cells exhibit cytokeratin-positive immunoreactivity in enzymatic staining.
Athymic nude mice (Nude Mouse, abbreviated as nude mice) have become an indispensable experimental animal model in the field of biomedical research. Through the efforts of experimental animal geneticists and breeders across the world, the nu gene has been introduced into different inbred animal strains, creating a series of animal models. For the mouse model alone, over 20 strains of inbred nude mice have been established. Due to their immunodeficiency, nude mice do not reject tissue grafts from heterologous animals under certain circumstances, making them ideal recipients for xenotransplantation of human malignant tumors. According to reports by T. Fogh et al. between 1979 and 1980, as many as 150 human tumor cell lines and primary human carcinomas have been successfully transplanted into nude mice. To date, human malignancies including colon cancer, breast cancer, lung cancer, ovarian cancer, melanoma, lymphoma, leukemia, renal cancer, cervical cancer, soft tissue sarcoma and osteosarcoma have been successfully transplanted into nude mice, with a certain percentage (35.7%) exhibiting favorable growth and being capable of serial passage. If established human tumor tissue culture cells are used as graft materials, the post-inoculation survival rate is higher (41%).
