This dataset contains whole-genome sequencing data of Neisseria gonorrhoeae isolates carrying the novel mosaic penA 34.019 allele, identified in Guangdong, China, between 2021 and 2023. The penA 34.019 variant harbors a unique G546S mutation that contributes to elevated resistance to extended-spectrum cephalosporins, including cefixime and ceftriaxone. These genomic data were generated to investigate the molecular characteristics, genetic background, and resistance mechanisms of penA 34.019-positive strains, which represent a newly emerging lineage associated with reduced susceptibility to cephalosporins. The dataset supports ongoing global surveillance efforts for antimicrobial-resistant N. gonorrhoeae.
