The study aimed to find out if there is any sequence preference in fly Argonaute loading. In HISSA, oligos were randomised at the 5'end and introduced into S2R+ FLAG-HA-AGO2 cell lysate for Argonaute loading. The HISSA libraries contain oligo-enriched reads.
