This study involved further evaluation of potential cancer therapeutics for Triple Negative Breast Cancer (TNBC), a highly metastatic and invasive cancer with limited treatment options. We examined in further detail the effect of the epigenetic modulator 4SC-202 on the transcriptome of TNBC tumors, formed in the highly metastatic 4T1 model. We hypothesized that 4SC-202 would reduce tumor burden and metastasis, possibly affecting the Cancer Stem Cell (CSC) population. 4T1 mouse TNBC cells were subcutaneously injected into the hind leg of female BALB/c mice, and mice were maintained as previously described (Messerl et al., 2019). The three groups were: 1) Control (vehicle); 2) 4SC-202; 3) the broad-spectrum HDAC inhibitor Vorinostat. Tumor growth was measured with calipers weekly and the lungs were evaluated for metastatic nodules after the termination of the experiment. RNA sequencing was performed on tumors isolated from the mice.
