Using the expression of IL-18 receptor (CD218) as a surface marker, CD69+ kidney-resident CD8 T cells can be separated into two subsets. Transcriptional analysis suggests that IL-18R low subset represents the mature tissue-resident T cells Overall design: 10000 congenically marked naïve P14 T cells were adoptively transferred into each sex-matched unmanipulated C57BL/6 mouse followed by LCMV Armstrong infection via an intraperitoneal route. 12 days after infection, lymphocytes were purified from the kidneys via enzymatic digestion and gradient centrifugation. Indicated P14 T cell subsets were FACS sorted from purified total kidney lymphocytes. Total RNA was extracted from freshly sorted cells. Kidneys from 10-15 recipient mice were pooled for each sorting.
