The purpose of the study was two-fold. First, to compare transcriptomic profiles of either MDA-MB-231 breast cancer cell lines or primary patient-derived xenograft (PDX) medulloblastoma cells grown in an engineered tissue constructs compared to standard 2D culture, static 3D culture, and orthotopic tumors. Second, to perform a large-scale CRISPRko screen of the kinome in the MDA-MB-231 cells, along with a targeted CRISPRko screen of drug-targetable genes in the PDX medulloblastoma cells in various growth conditions. Orthotopic tumors were located in either the mouse mammary fat pad (for MDA-MB-231 cells) or the mouse cerebellum (for PDX medulloblastoma cells). 2D culture was either on a standard tissue culture plate (for MDA-MB-231 cells) or in suspension culture (for PDX medulloblastoma cells).
