Immune checkpoint blockade (ICB) has opened a new era in cancer therapy. Here, we studied the potential role of eosinophils in the response to ICB treatment and chemotherapy combination in mouse models of primary and metastatic breast cancer. As a model of metastatic breast cancer, we used mice with metastasis originating from orthotopically transplanted mammary tumors, which in turn derived from spontaneous mammary tumors of K14cre;Cdh1F/F;Trp53F/F mice. In this study, the gene expression profile of circulating eosinophils sorted from mice with metastatic cancer treated with a combination of cisplatin and anti-PD1 plus anti-CTLA-4 antibodies was analyzed br RNA-sequencing. Comparative gene expression profiling analysis of RNA-seq data for mouse eosinophils sorted from blood of mice with metastatic cancer treated with cisplatin + anti-PD1 + anti-CTLA4 antibody (n=7 mice) or with control antibody alone (n=4 mice).
