Pancreatic stellate cells can be activated to a myofibroblast phenotype in response to TGFB treatment. The goal of this study was to determine how inhibition of PIN1 affects stellate cell state at baseline and in response to TGFB treatment. Overall design: Primary human pancreatic stellate cells (PSCs) were cultured in Stellate Cell Media. For inhibitor studies, cells were pre-treated for 48 hours with 10 uM all trans retinoic acid (ATRA), 10 uM KPT-6566 (KPT), or DMSO as a control. Media was then changed to replenish this treatment as well as to treat with and without 5 ng/mL TGFB1. Cells were collected 24 hours later, RNA isolated and used for RNA sequencing.
