We performed Sleeping Beauty transposon based insertional mutagenesis screening in mice and identified more than 1000 candidate colorectal cancer genes. By compareing to the dataset for genes mutated in human colorectal cancer, we were able to enrich genes commonly mutated between human and mice. In this project, we set up the experimental system to validate these candidate cancer driver genes using CRISPR-Cas9 system and intestinal organoids culture system.
