Thymosin a1 Inhibiting Ferroptosis of Dental Pulp Cells in Pulpitis

Tooth pulpitis is a common oral ailment. Understanding the underlying mechanisms of pulpitis and developing effective treatment strategies are of significant importance.In this study, single-cell RNA sequencing (scRNA-seq) was used to identify cellular heterogeneity between 3 pulpitis tissue and 3 healthy pulp tissue. Raw data of ScRNA-seq of 3 healthy pulp tissue have been uploaded (GEO accession: GSE274562; Control1: GSM8452929; Control2: GSM8452930; Control3: GSM8452931). Overall design: To dissect cellular heterogeneity and investigate ferroptosis in each type of cells in inflammatory dental pulp, we harvested 3 healthy dental pulp and 3 inflammatory dental pulp. The 6 samples were dissociated into single cells suspension and then performed scRNA-seq. After quality control, the transcriptomes of 40,231 cells (Pulpitis: 17,814; Healthy pulp: 22,417) were obtained. These cells were subsequently visualized into 12 distinct cell clusters including CD4+ T cell, CD8+ T cell, B cell, Macrophage, Neutrophil, Mast cell, Plasma cell, Fibroblast, Endothelial cell, Mesenchymal stem cell (MSC), Schwann cell (ScC), nonmyelinating ScC (nmScC), by generating uniform manifold approximation and projection (UMAP) plots, based on hierarchical clustering and established lineage-specific marker genes
