Independent first-principles rediscovery of RB1-E2F synthetic lethality on the Faure 2006 cell-cycle Boolean network in 41.5 ms

The Faure et al. 2006 mammalian cell-cycle Boolean network (10 nodes, published in J Theor Biol 246:723), when scanned by a generic pair-knockout brute-force attractor enumerator, rediscovers E2F as the canonical RB1-loss synthetic-lethal partner in 41.5 ms of total wall time. E2F is the textbook RB1-loss SL target; E2F-axis inhibitors (BET-bromodomain, CDK7, CDK9 family) are in active clinical development for RB1-loss SCLC and triple-negative breast cancer subsets.
