454 pyrosequencing analysis of bacterial communities in human atherosclerotic plaques

Atherosclerotic plaques were removed by endarterectomy from asymptomatic and symptomatic patients. The plaques were divided into three regions known to display different histological vulnerability: A, upstream of maximum stenosis; B, site of maximum stenosis; C: downstream of maximum stenosis. To determine bacterial composition, DNA was isolated from each plaque region and 16S rRNA genes were amplified with PCR, followed by 454 pyrosequencing.
