We used Affymetrix Mouse Exon 1.0 ST arrays to identify potential changes in alternative splicing between a subcongenic interval of the QTL Sicd2 on Chr 15 and their FVB-like littermates. Results implicate novel candidate genes conferring susceptibility to seizure-induced cell death. Total RNA was extracted from the hippocampus of 8 Sicd2-ISCL4 subcongenic mice and 8 FVB-like littermates.
本研究采用Affymetrix小鼠外显子1.0 ST基因芯片(Mouse Exon 1.0 ST arrays),对15号染色体上数量性状位点(Quantitative Trait Locus, QTL)Sicd2的亚同源导入系区间小鼠,与其同窝FVB品系相似仔鼠之间的可变剪接(alternative splicing)潜在变化进行了鉴定。研究结果揭示了若干可赋予惊厥诱导细胞死亡(seizure-induced cell death)易感性的新型候选基因。本研究从8只Sicd2-ISCL4亚同源导入系小鼠及8只同窝FVB品系相似仔鼠的海马体(hippocampus)中提取了总RNA(Total RNA)。
