采集稻田土-水界面的周丛生物膜到实验室中进行不同磷素浓度的室内模拟培养。分别设置高、中、低三个磷素浓度,并且磷素浓度为唯一变量及限制因子。细胞在人工气候室中进行25℃,12小时光照/天的培养。每个处理设置4个重复。培养箱长30cm宽20cm。于细胞生长对数期采集细胞培养液样品。主要步骤如下:轻微摇晃培养箱,用注射器抽取100ml培养液到离心管中,经过离心,过0.22微米滤膜后,获得周丛生物膜的胞外代谢物样品,共12个样品。后续进行胞外代谢物的总浓度分析。经过除盐和浓缩处理后,上机FT-ICR MS获得胞外代谢产物的物质组成及元素组成数据。
Periphytic biofilms collected from the soil-water interface of paddy fields were transferred to the laboratory for indoor simulated cultivation with varying phosphorus (P) concentrations. Three phosphorus concentration levels (high, medium, low) were established, with phosphorus concentration serving as the sole variable and limiting factor. The cultivation was conducted in an artificial climate chamber at 25°C with a 12-hour light period per day. Four replicates were set for each treatment. The incubation vessels measured 30 cm in length and 20 cm in width. Samples of the cell culture medium were collected during the logarithmic growth phase of the biofilm cells. The main procedures are as follows: Gently shake the incubation vessels, withdraw 100 mL of the culture medium into a centrifuge tube using a syringe, followed by centrifugation and filtration through a 0.22 μm microporous membrane to obtain extracellular metabolite samples of the periphytic biofilms, with a total of 12 samples. Subsequent analysis of the total concentration of extracellular metabolites was carried out. After desalination and concentration processing, Fourier Transform Ion Cyclotron Resonance Mass Spectrometry (FT-ICR MS) was employed to acquire data on the molecular composition and elemental composition of the extracellular metabolites.
