ChIP-seq identified the regulation of genes by the quorum-sensing regulator OpaR in Vibrio parahaemolyticus

We report that ChIP-seq analyses of the quorum-sensing regulator OpaR at late-log and stationary phase in Vibrio parahaemolyticus Overall design: We inserted a FLAG-tag before the opaR gene on chromosome. FLAG-OpaR/VP93 was grown to a late-log phase (OD600 3) and stationary phase (OD600 4.2). DNA fragments were sheared by Bioruptor Pico (Diagenode). Anti-FLAG antibody beads (Sigma) were used to enrich the IP DNA fragments. ChIP DNA was then purified using the Agencourt AMPure XP PCR Purification Kit (Beckman Coulter).
