This study was designed to investigate the effect of PCR primers that include an index sequence. DNA was extracted from the retentate of 1L seawater samples (n = 11) over 0.45 micron filter membranes. Amplicons of the mitochondrial 16S region were generated using either a single PCR with indexed primers, or with two PCRs, first with non-indexed primers, and subsequently with indexed primers.
