This dataset comprises the experimental data supporting the functional analysis of the SSU0990 gene in Streptococcus suis. The study investigates the phenotypic and molecular consequences of SSU0990 gene deletion and point mutations.<br>The contents include:<br>Molecular Validation and Protein Work:<br>PCR verification data confirming the successful deletion of the SSU0990 gene from the S. suis genome.<br>Data related to the large-scale expression and purification of the protein encoded by the SSU0990 gene, both in its wild-type form and after introducing point mutations.<br>Phenotypic Characterization of the Mutant Strain:<br>Growth Kinetics (Fig. 2B-E): Growth curve data (OD600 measurements) for the wild-type (P1/7), SSU0990 deletion mutant (ΔSSU0990), and complementation strain (CΔSSU0990). The dataset also includes calculated parameters of lag time, maximum growth rate, and generation time.<br>Stress Susceptibility (Fig. 3A-E): Data on the sensitivity of the strains to various stressors, presented as inhibition zones for hydrogen peroxide (H2O2), penicillin (PEN), rifampicin (RIF), and ciprofloxacin (CIP). It also includes measurements of Tail DNA%, indicative of DNA damage.<br>Mutation Analysis (Fig. 4A-D): Data on spontaneous and induced (H2O2) mutation frequencies for the three strains. Additionally, the spectrum of spontaneous mutation types (e.g., A>T, A>G) is provided.<br>Enzymatic Function Analysis (Fig. 5E-G):<br>Biochemical assay data characterizing the phosphatase activity of the purified SSU0990 protein (wild-type and mutant variants) against oxidized guanine nucleotides (8-oxo-dGTP, 8-oxo-dGDP, 8-oxo-dGMP). This includes time-dependent and concentration-dependent hydrolysis curves.<br>Virulence Assessment (Fig. 6A-E):<br>Data from infection models showing the phagocytosis rate, survival rate, invasion rate, and bacterial load in the brain for the wild-type, mutant, and complementation strains, demonstrating the role of SSU0990 in virulence.<br>Overall, this dataset provides evidence that the SSU0990 gene plays a critical role in the oxidative stress response, DNA damage repair, and full virulence of Streptococcus suis. The encoded protein exhibits phosphatase activity, suggesting a mechanism for its function in sanitizing the nucleotide pool.
