Cells from porcine lymphoma tumors in the lymph node and spleen were cultured, cloned, and sequenced using Illumina next generation sequencing and compared to CD21+ or CD3+ splenocytes. Lymphoma clones were identified a B cell lymphomas by gene expression and surface marker presence. Overall design: Cells from 4 different lymphoma clones from the subileac lymph node, 2 lymphoma clones from the spleen, a CD21+ splenocyte sample to be used as a normal B cell control, and a CD3+ splenocyte sample to be used as a normal T cell control were used for RNA isolation and paired end sequencing on an Illumina HiSeq 2500. Raw data was analyzed at the University of Minnesota Informatics Institute and aligned to the Sus Scrofa11.1 genome. The lymphoma sequences were compared to the B cell and T cell controls separately to determine if they were more similar to B or T cells. Since the lymphoma cells were more similar to B cells, they were then compared against the CD21+ B cell control to identify differentially expressed genes between control B cells and the lymphoma cells.
