A proposed model to summarize the ability of GITR to modulate TNF-α induced canonical NF-κB pathway. The presence of GITR in MM cells could disrupt TNF-α/TNFR induced NF-κB activation by blocking phosphorylation of IKK-beta, subsequenctly leading to early termination of nuclear translocation of NF-κB p65/p50 and impaired induction of BCL-2 and Survivin.
