To investigate the functional role of SLC7A7 in bladder cancer biology, SW780 human bladder carcinoma cells were stably transfected with SLC7A7-targeting lentiviral vectors (SW780-shSLC7A7) using lentivirus-mediated RNA interference technology. Parallel control cell lines (SW780-shNC) were generated through stable transfection with non-targeting scrambled shRNA vectors to establish appropriate negative controls. Following successful cell line establishment, extracellular vesicles were isolated from conditioned culture media of both SW780-shSLC7A7 and SW780-shNC cell populations using differential ultracentrifugation coupled with density gradient purification. The purified exosomal fractions were subsequently subjected to small RNA sequencing analysis (Illumina NovaSeq 6000 platform) to systematically profile miRNA expression patterns and identify differentially expressed exosomal miRNAs associated with SLC7A7 knockdown. This comparative exosomal miRNA profiling approach aims to elucidate potential extracellular communication mediators involved in SLC7A7-mediated oncogenic pathways.
