Bulk RNA sequencing analysis of human airway basal stem cells stimulated by BALF from mouse fibrotic lungs and normal lungs

To understand how the microenvironment of fibrotic lungs affects the behavior of airway basal stem cells (BCs), we constructed a mouse model of pulmonary fibrosis using bleomycin induced injury, and collected bronchoalveolar lavage fluid (BALF) from the control group and bleomycin damaged mice on day 14 after injury. Then, human BCs were stimulated with control or bleomycin BALF in culture and subjected to bulk RNA sequencing. Transcriptomic analysis showed that basal stem cells stimulated with bleomycin BALF upregulated genes related to chemotaxis and migration, and activated the TGF-Ã pathway. Overall design: Human BCs were stimulated with PBS or bleomycin BALF in culture for 48 hours and subjected to bulk RNA sequencing. Total RNA was extracted from Human basal cells using Trizol (15596018CN, Invitrogen), and RNA extraction was performed according to the manufacturer's instructions. Subsequently, the extracted RNA was treated with Dnase I (Invitrogen, Life Technologies, USA) to remove any contaminating DNA. Utilize the Novogene platform for cDNA library construction and sequencing.
